Differentiation of Mesenchymal Stem Cell toward the Insulin-like Cells with Lentivirus Vector Mir-375

چکیده مقاله:
Background & Objective: Type1 diabetes is characterized by autoimmune destruction of pancreatic β cells, leading to reduced insulin secretion. Differentiation of mesenchymal stem cells (MSCs) into β-like cells offers new ways of diabetes treatment. MSCs can be insulated from the human umbilical cord tissue and differentiate into insulin-producing cells. Material & Methods: Human umbilical cord–derived stem cells (hUDSCs) were attained after birth, selected by plastic adhesion, and considered by flow cytometric analysis. For the differentiation induction, miR-375 was lentivirally over expressed in hUDSCs. Results: Meaningful that microRNAs (miRNAs) are the key players in several stages of pancreatic Differentiation, we have presented a novel and cost-effective strategy in which the over-expression of miR-375 promotes the pancreatic differentiation in hUDSCs in the absence of any other stimulator. The major expressions of PDX1 and insulin were identified by quantitative Reverse Transcription Polymerase Chain Reaction (RT-PCR) (P <0.05). Conclusion: In conclusion, the Morphological assessment and the expression analysis of islet marker genes demonstrated that hUDSCs are able to differentiate into insulin-producing cells by transduction with lentiviral vector miR-375

Immunoregulatory Effects of Glutathione During Mesenchymal Stem Cell Differentiation to Hepatocyte-Like Cells
Background: The role of mesenchymal stem cell in cellular therapy is the subject of interest for many researchers. The differentiation potential of MSCs and abilities in modulations of the recipient’s immune system makes them important cells in tissue regenerative studies. MSCs by releasing the proinflammatory cytokines play important role in immunomodulatory systems; however the signaling path...

Objective(s):To culture thein vitro mouse embryonic stem cells (mESCs) and to direct their differentiation to germ-line cells; in present study we used a vector backbone containing the fusion construct Stra8-EGFP to select differentiated ES cells that entered meiosis. Retinoic acid was used to differentiate embryonic stem cells to germ cells. Materials and Methods: A fragment of Stra8 gene pr...
متن کاملPromotion Effects of miR-375 on the Osteogenic Differentiation of Human Adipose-Derived Mesenchymal Stem Cells
MicroRNA plays an important role in bone tissue engineering; however, its role and function in osteogenic differentiation warrant further investigation. In this study, we demonstrated that miR-375 was upregulated during the osteogenic differentiation of human adipose-derived mesenchymal stem cells (hASCs). Overexpression of miR-375 significantly enhanced hASCs osteogenesis both in vitro and in ...
متن کاملThe Generation of Insulin Producing Cells from Human Mesenchymal Stem Cells by MiR-375 and Anti-MiR-9
BACKGROUND MicroRNAs (miRNAs) are a group of endogenous small non-coding RNAs that regulate gene expression at the post-transcriptional level. A number of studies have led to the notion that some miRNAs have key roles in control of pancreatic islet development and insulin secretion. Based on some studies on miRNAs pattern, the researchers in this paper investigated the pancreatic differentiatio...
متن کاملIn-vitro Differentiation of Human Umbilical Cord Wharton’s Jelly Mesenchymal Stem Cells to Insulin-Producing Cells
Background & Objective: Diabetes is a major chronic metabolic disease in the world. Islet transplantation is a way to treat diabetes. Unfortunately, this method is restricted due to graft rejection and lack of donor islets. Mesenchymal Stem Cells (MSCS) have the ability to differentiate into Insulin-Producing Cells (IPCs). In this study, Human Umbilical Mesenchymal Stem Cells (HUMSCS) were in...
